You can draw them directly into Anime Studio on a vector layer. Or you can draw them in another program and export a png with a transparent background. Do you need Anime Studio to interpolate the transition between the mouth positions? Or is it okay if the mouths flash from one position to the next? That will make a big difference in the complexity of your mouths. In my opinion, making mouths that do not need to interpolate is a lot easier. But so...
