Just to add my 2 cents ...
The only time I have had similar is when another Windows app like Audition or Studio One is hogging the audio device. But in those cases the audio won't play in WMP, Groove, VLC etc. either.

Oh, yeah...I ran into that issue just last week during a work meeting; nobody on the call could hear me even though my computer and software was clearly 'unmuted'. It took me a minute to realize that an audio program I had been using the night before was hijacking audio control. After I shut it down, and reconnected to the call, the audio was fine.
