Does a bone's "direction" make a difference?

Does it make a difference as to the direction a bone's pointing toward? If it does, in what way?

When you rotate the bone, it will rotate around the origin (the point the bone is pointing from) and the other end of the bone will swing around. Is this what you're asking?
