Try the Fill Quad Mesh script in the Script Draw menu. Besides creating valid quad shapes, this tool also flags the problems with your drawing to help you fix it. It's super useful.
Tip: Use Sharp Corners and Auto-weld when drawing your quad mesh. Also, remember you can combine quads with triangles...this is especially useful in face setups around the eyes and mouth.
Tip #2: In Moho 13.5, Select Shape finally has a Select All shapes button. Use this when you need to make all your mesh shapes have the same style.

Re: How do I use the quad mesh feature?
Also, if all you're wanting at this point is to try out the facility, the quickest way to make a quad mesh is from the Draw Shape tool (standard shortcut S) and pick the Grid option - best to use autoFill only.
If you draw your own grid, as Greenlaw says, the Fill Mesh tool will pick up on anything that isn't a valid 3- or 4-sided shape (it will also make all the points "sharp corners") - common problems are two points with two paths between them (i.e. a shape with just two points); points within a pixel of an unconnected shape (i.e. visually the shapes have merged even though they're separate) ; or 5+ sided shapes - without seeing what you've drawn, impossible to tell... if you want to upload what you have, we can take a look.
with a valid mesh, choose the vector (or image) layer you want it to control, and in the "vectors" (or "image") tab, select the grid as the "Smart Warp Layer". On frame 1 onwards, point motion in the grid will warp the controlled layer.
