You're describing a Boolean operation. There are no Boolean operations in AS -- it's a (mostly) two-dimensional animation program...
You can't have everything. Where would you put it?

Remove the color from both objects, using the "delete shape"-tool (in the box's case, just from the light purple part). Now select both the square and the circle.
When you select the "create shape"-tool, you should already see that Moho treats the circle as a hole in the object. Just add the desired color, and there ya go.
