How Do I work ( Fade in & Fade out) in anime studio

You can animate a layer's alpha setting (opacity) from 0 top 100% to fade in, and from 100% to 0 to fade out. Or you could fade to and from a totally black layer to fade in or out the same way.

Read the built-in Anime Studio Users Manual, section User Interface->Layers Window and search for Layer Settings. There you'll find the "Opacity" parameter of the layer (commonly known as "alpha" for raster image files). Its default value is 100% (opaque) and can be set to 0% (fully transparent)

Reading the built in Users Manual is a good and worthly task to fully understand AS's features.
